Production of plasma esterified cholesterol in lean, normotriglyceridemic humans.

چکیده
The rate of production of plasma esterified cholesterol was measured both in vivo and in vitro in seven subjects and in vivo alone in eight subjects. All subjects were lean, clinically healthy, and had triglyceride concentrations less than 1.5 micro moles/ml. In vivo production was calculated from the labeling of free and esterified cholesterol in plasma samples collected at 1-hr intervals for 8 hr after an intravenous injection of [(3)H]mevalonic acid, on the assumption that plasma free cholesterol was the sole immediate precursor of esterified cholesterol. In vitro production was measured in serum samples collected 1 hr after the injection of [(3)H]mevalonic acid (when radioactivity in esterified cholesterol was very low relative to that in free cholesterol); these samples were incubated for 1 hr at 37 degrees C. The rates measured in vivo and in vitro were very similar in the seven subjects, strengthening the confidence in the techniques. In vivo production was measured during the postabsorptive state in all 15 subjects and in 5 of them also during the last 8 hr of a 32-56-hr period when all calories were taken in three hourly meals of an 80% carbohydrate, fat-free formula. In the postabsorptive state there was no apparent relationship between the production of esterified cholesterol and the concentration of either free or esterified cholesterol. Rather, despite a wide range of cholesterol concentrations, esterified cholesterol production was similar in all subjects. During the carbohydrate consumption the esterified cholesterol concentrations were significantly lower than during the postabsorptive state, but there was virtually no change in rate of production. It has been concluded that the differences in concentration of esterified cholesterol in lean, normotriglyceridemic subjects cannot be explained solely on the basis of differences in its production.

Studies on the turnover of triglyceride and esterified cholesterol in subjects with the nephrotic syndrome.
Factors involved in the hyperlipidemia of nephrosis have been studied in seven patients. The turnover of triglyceride was measured in plasma very low density lipoproteins after the injection of glycerol-(14)C. The turnover of esterified cholesterol was measured in whole plasma and in very low density lipoproteins after the injection of mevalonic acid-2-(3)H. Urine protein loss was found to be s...
متن کاملReduced atherogenesis in cholesterol-fed diabetic rabbits. Giant lipoproteins do not enter the arterial wall.
In cholesterol-fed rabbits, alloxan-diabetes has an anti-atherogenic effect, which is associated with severe elevation of plasma triglyceride concentrations. To study this effect, we measured lipoprotein sizes and aortic permeability coefficients for cholesteryl ester and for albumin in hypertriglyceridemic diabetic cholesterol-fed rabbits and in normotriglyceridemic cholesterol-fed rabbits. Wi...
متن کاملDyslipidemia in obese cats.
Obesity is an important endocrine disorder in cats and is a risk factor for diabetes similar to humans. The goal of this study was to examine the effect of long-term obesity and different diets (high protein, and high carbohydrate supplemented with saturated fatty acids or n-3 polyunsaturated fatty acids) on plasma lipids in the fasted and fed states in 12 lean (LEAN) and 12 obese (OBESE) cats ...
متن کاملObservations concerning the production and excretion of cholesterol in mammals; I. Plasma cholesterol after bile duct ligation and free cholesterol injection.
Investigators (l-6) are agreed that in man, the dog, and the rat biliary obstruction is followed by a rise in the total cholesterol concentration of the blood. Opinion is not uniform concerning the changes in blood content of esterified cholesterol after biliary obstruction. Thus, Epstein and Greenspan (3,4) observed a rise in cholesterol ester concentration of the blood in the majority of thei...
متن کاملLipoprotein cholesteryl ester production, transfer, and output in vivo in humans.
Our aim was to identify and quantify the major in vivo pathways of lipoprotein cholesteryl ester transport in humans. Normal (n = 7), bile fistula (n = 5), and familial hypercholesterolemia (FH; n = 1) subjects were studied. Each received isotopic free cholesterol in HDL, LDL, or particulate form, along with another isotope of free or esterified cholesterol or mevalonic acid. VLDL, intermediate...
